TRPA1 Agonists and Bladder Nociception in Female Rats Suggest Potential for Nutraceutical Benefit from Cinnamon

چکیده

TRPA1-related drugs alter sensation, particularly in conditions of inflammation. To further characterize the role these bladder TRPA1 agonist cinnamaldehyde (CMA) and oral true cinnamon spice were examined preclinical models pain. Female adult rats, with without acute zymosan-induced cystitis, anesthetized visceromotor (VMR) cystometric responses to urinary distension (UBD) determined following either intravesical administration CMA/vehicle solutions or cinnamon/vehicle. ELISA measures content also determined. Acute cystitis resulted increases produced an increased vigor VMRs UBD a lowering micturition volume thresholds for activation response. Intravesical CMA robust inhibition rats but not those without. Micturition lowered by had no additional effect cystitis. Oral mild augmentation A potentially analgesic spice, cinnamon, treatment pain was suggested studies. Human studies are indicated.

TrpA1 Regulates Thermal Nociception in Drosophila

Pain is a significant medical concern and represents a major unmet clinical need. The ability to perceive and react to tissue-damaging stimuli is essential in order to maintain bodily integrity in the face of environmental danger. To prevent damage the systems that detect noxious stimuli are therefore under strict evolutionary pressure. We developed a high-throughput behavioral method to identi...

Annexin A2 regulates TRPA1-dependent nociception.

The transient receptor potential A1 (TRPA1) channel is essential for vertebrate pain. Even though TRPA1 activation by ligands has been studied extensively, the molecular machinery regulating TRPA1 is only poorly understood. Using an unbiased proteomics-based approach we uncovered the physical association of Annexin A2 (AnxA2) with native TRPA1 in mouse sensory neurons. AnxA2 is enriched in a su...

Prostaglandin metabolite induces inhibition of TRPA1 and channel-dependent nociception

BACKGROUND The Transient Receptor Potential (TRP) ion channel TRPA1 is a key player in pain pathways. Irritant chemicals activate ion channel TRPA1 via covalent modification of N-terminal cysteines. We and others have shown that 15-Deoxy-Δ12, 14-prostaglandin J2 (15d-PGJ2) similarly activates TRPA1 and causes channel-dependent nociception. Paradoxically, 15d-PGJ2 can also be anti-nociceptive in...

GLT-1 overexpression attenuates bladder nociception and local/cross-organ sensitization of bladder nociception.

Glutamatergic pathways mediate transmission of pain. Strategies to reduce glutamatergic neurotransmission may have beneficial effects to mitigate nociception. Recent work revealed that overexpression of the astrocytic glutamate transporter (GLT-1) by transgenic or pharmacologic approaches produced a diminished visceral nociceptive response to colonic distension. The purpose of this study was to...

Fetal ethanol exposure attenuates aversive oral effects of TrpV1, but not TrpA1 agonists in rats.

In humans, fetal ethanol exposure is highly predictive of adolescent ethanol use and abuse. Prior work in our labs indicated that fetal ethanol exposure results in stimulus-induced chemosensory plasticity in the taste and olfactory systems of adolescent rats. In particular, we found that increased ethanol acceptability could be attributed, in part, to an attenuated aversion to ethanol's aversiv...
